Firstly, P21 aka cyclin-dependent kinase inhibitor 1 or CDK-interacting protein 1, is a cyclin-dependent kinase inhibitor (CKI). Another words, a nootropic peptide targeting improved neurogenesis. In animal studies, it shows to boost levels of BDNF (Brain-derived neurotrophic factor), thus promoting neurogenesis (nerve growth) and inhibiting the formation of Amyloid plaques and Tau proteins seen in Alzheimer’s disease.
How P21 works:
Secondly, P21 works by enhancing neurogenesis in the dentate gyrus. The dentate gyrus is part of the hippocampal formation and plays a role in the formation of new episodic memories as well as the exploration and learning that happens in new environments and situations. This part of the brain is also responsible for pre-processing information and promoting pattern separation which enables mammals to differentiate memories.
According to research, it can induce a variety of effects in the body. This includes heightened BDNF (Brain-derived neurotrophic factor) levels, enhanced phosphorylated GSK3B (Glycogen Synthase Kinase 3 Beta: a Protein Coding gene) levels, and increased AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid) receptor protein levels.
Research in mice indicates that P21 doesn’t bind to the CNTF receptor, implying that it is not an analogue of CNTF (Ciliary neurotrophic factor). Instead, it seems to inhibit antibodies and molecules that neutralize it. Subsequently, P21 increases the concentration of CNTF and mimics its effects.
Further research suggests that its levels also increases BrdU (Bromodeoxyuridine / 5-bromo-2′-deoxyuridine) positive cells (synthetic nucleosides) in the dentate gyrus. In the same study, researchers found that P21 also promotes cell proliferation in this region of the dentate gyrus, supporting the theory that this proliferation is actually strengthening neurogenesis.
